The town is located in Hillsborough County, adjacent to Greenville, Hollis, Milford, Nashua, and the Massachusetts towns of Townsend and Pepperell. Brookline is about 30 minutes from Manchester.

At the last census (2010), its population was about 4,990. According to city-data.com, Brookline’s estimated median household income in 2012 was $99,750, and its estimated median home value was $306,138.

Brookline’s two elementary schools form the Brookline School District, while middle- and high-school students attend schools with Hollis students in the Hollis-Brookline Cooperative School District.
